Mutation of a cystein residue
C124 to serine (C124S) renders the PTEN protein unable to perform
its phosphatase activity. wild-type PTEN and the C124S PTEN mutant
were both tagged with His-Ni and purified. They were each
independently incubated in vitro with an equal amount of histones
for detection of PTEN-bound histone H1. The result of the
co-precipitation is presented in the figure below. Is the
phosphatase activity of PTEN necessary for its interaction with
